基于酶活性丹参茎叶散剂活血活性谱—效模型研究

摘 要:采用HPLC法和纤维蛋白原平板法,测定6批丹参茎叶散剂的特征图谱和体外活血活性值,并对特征图谱的特征峰进行指认,分析其特征峰成分与活血活性的相关性;采用SPSS软件,经主成分分析、相关性分析和多重线性回归分析的方法构建其谱—效模型,并用6批丹参茎叶散剂对其进行验证.丹参茎叶散剂特征图谱中得到9个特征峰,体外活血活性值在132.52~153.25 U/mL.根据主成分分析,综合解释总方差和碎石图分析,得该模型的最优主成分数为5,由Spearman相关性分析得到5个主成分色谱峰和活血活性相关性均大于0.657,且对活血作用显示正相关,预测值和测量值的相对误差较小.建立的基于酶活性的活血活性谱—效模型可较好地预测丹参茎叶散剂的活血活性,为其进一步开发利用提供基础.

Study on Activity Spectrum Effect Model of Stem and Leaf-Powder of Salvia Miltiorrhiza Based on Enzyme Activity

Abstract:Six batches of Salvia miltiorrhiza stem and leaf powder were determined by HPLC and fibrinogen plate method.The feature map and in vitro activating activity value of Salvia miltiorrhiza stem and leaf powder were measured.The characteristic peak of the feature map was identified by reference substance,and the correlation between the component of the characteristic peak and its activating activity was analyzed.By using the SPSS software,the spectral efficiency model was established by principal component analysis,correlation analysis and multiple linear regression analysis method,and 6 batches of Salvia miltiorrhiza stem and leaf powder were used for verification.The results showed that nine characteristic peaks were obtained in the feature map of stem and leaf powder of Salvia miltiorrhiza.The activating blood activity value in vitro was 132.52 ~ 153.25 U/mL.According to principal component analysis,comprehensive interpretation of total variance and gravel diagram analysis,the optimalprincipal component number of this model was 5.Spearman correlation analysis showed that the correlation between the five principal component chromatographic peaks and the activating blood activity was all greater than 0.657,and showed a positive correlation with the activating blood activity.The relative error between the predicted value and the measured value was small.It is concluded that the active blood activity spectrum effect model based on enzyme activity can predict the active blood activity of the stem and leaf powder of Salvia miltiorrhiza,and provide the basis for its clinical application.
